Coexpression of NG2, neural/glial antigen 2, in the developing murine brain is regulated by TGF-beta.
The study of neural/glial antigen 2 (NG2), an integral membrane proteoglycan, has shown that this molecule is closely associated with the growth and migration of neuroblasts during the early development of the central nervous system. In order to assess the influence of TGF-beta during the development of this proteoglycan, we have produced a NG2-specific monoclonal antibody that has been used to study the expression of this molecule in the developing murine brain. Here we report that NG2 is strongly expressed in migratory neuroblasts from embryonic day (E) 12.5 to E15.5 in both the embryonic and the extraembryonic neural tube, whereas postnatal NG2 expression is restricted to glial cells of the cortex and other regions of the CNS. In addition, NG2 is transiently expressed in radial glial cells of the developing ventricular wall from E14.5 to E15.5. The results indicate that TGF-beta is a major regulator of NG2 expression in the developing CNS. The importance of this regulatory molecule is emphasized by the observation that both NG2 protein and mRNA are strongly expressed in developing glial cells of the cortex from E14.5 to E17.5 in TGF-beta null embryos.
